Untapped Potential for Cardiac Repair

چکیده
An explosion of interest in new therapies for heart repair has occurred recently. Most notably, clinical trials of cell-based cardiovascular therapy have garnered much attention, and preliminary results are encouraging. However, the mechanism of action of cell-based therapies remains unclear, and many details with regard to characterization, quality control, and delivery of cells remain to be worked out before widespread therapeutic application. Nonetheless, the reported efficacy of cardiac cell–based therapies in human trials has generated interest in additional therapeutic development pathways. The identification of the release of paracrine-acting proteins as one mechanism by which cell-based therapies in the heart may improve cardiac function has spurred renewed interest in protein-based therapeutic approaches for cardiac repair. Although protein therapeutics are increasingly becoming mainstream in a number of fields, including cancer and inflammatory diseases, (9 of the 20 top-selling drugs in 2012 were proteins) proteins have relatively low penetration in the cardiovascular market. This is not to suggest a lack of progress in the cardiovascular therapeutic development, because smallmolecule drugs such as statins and anticoagulants have been huge successes. Rather, as overall new drug approvals shift predominantly to proteins, protein therapeutic development represents an area of opportunity for cardiovascular medicine. Mesenchymal stem cells from umbilical cord tissue as potential therapeutics for cardiomyodegenerative diseases – a review
Heart failure is one of the leading causes of death worldwide. End stage disease often requires heart transplantation, which is hampered by donor organ shortage. Tissue engineering represents a promising alternative approach for cardiac repair. For the generation of artificial heart muscle tissue several cell types, scaffold materials and bioreactor designs are under investigation. In this revi...
متن کاملPerivascular cells and tissue engineering: Current applications and untapped potential
The recent development of tissue engineering provides exciting new perspectives for the replacement of failing organs and the repair of damaged tissues. Perivascular cells, including vascular smooth muscle cells, pericytes and other tissue specific populations residing around blood vessels, have been isolated from many organs and are known to participate to the in situ repair process and angiog...
متن کاملTherapeutic angiogenesis promotes efficacy of human umbilical cord matrix stem cell transplantation in cardiac repair
Objective(s):Although previous studies have confirmed the beneficial effects of human umbilical cord matrix stem cell (hUCM) transplantation post myocardial infarction (MI), but this stem cell resource has no potential to induce angiogenesis. In order to achieve the process of angiogenesis and cardiomyocyte regeneration, two required factors for cardiac repair agents were examined namely; hUCM ...

It was assumed that the loss of cardiomyocytes is irreversible. The main goal is to develop widely available and clinically applicable treatments for heart diseases. The several studies have showed that the use of stem cells can improve complicacies such as cardiovascular diseases. Stem cells have a potential benefit of the self-renewal and cell differentiation into the cell types that can play...
متن کاملAre Stem Cells the next Therapeutic Tool for Heart Repair?
Cardiovascular disease remains the leading cause of morbidity and mortality in the United States and Europe. In recent years, the understanding that regenerative processes exist at the level of the myocardium, has placed stem cell research at center stage in cardiology. A stem cell is a cell that has the ability to divide (self replicate) for indefinite periods often throughout the life of the ...
